The method uses two dyes in sequence:
1. The May Grünwald solution, consisting of methylene blue eosinate that colors the nuclei in blue and the basophilic cytoplasm in red-pink.
2. The Giemsa solution, complex mixture, consisting of methylene blue chloride, methylene blue eosinate, sky-blue II eosinate, which increases the intensity of the nuclear staining and the ability to selectively highlight the cell elements.
